PL-02 Vernalization – Cold-mediated epigenetic regulation of a developmental switch

چکیده

At a certain stage in their life-cycle plants flower, that is they undergo the transition from vegetative to reproductive development. The correct timing of this transition is crucial for reproductive success, so the plant integrates multiple environmental and endogenous signals to judge when to flower. The Dean laboratory is studying the importance of prolonged cold for flowering, a process known as vernalization. Our current view of the vernalization pathway has been built up though identification of Arabidopsis mutants and analysis of the corresponding genes, complemented by chromatin biochemistry. The vernalization pathway in Arabidopsis mediates the silencing of a target – FLC, by a conserved Polycomb mechanism. The talk will describe our understanding of the triggering of FLC repression by prolonged cold; the nucleation of chromatin silencing at a specific site in FLC; and the spreading of the silencing yet spatial restriction to FLC. We are aiming to describe this epigenetic silencing in a quantitative model and our efforts to date will be presented. Arabidopsis accessions have adapted to a wide range of latitudes and climates. We are studying whether variation in vernalization has contributed to this adaptation and have found that molecular variation at FLC is an important component. Our current understanding of this variation, its evolutionary origins and ecological significance will be discussed.

Vernalization-mediated epigenetic silencing by a long intronic noncoding RNA.

Vernalization is an environmentally-induced epigenetic switch in which winter cold triggers epigenetic silencing of floral repressors and thus provides competence to flower in spring. In Arabidopsis, winter cold triggers enrichment of tri-methylated histone H3 Lys(27) at chromatin of the floral repressor, FLOWERING LOCUS C (FLC), and results in epigenetically stable repression of FLC. This epig...

Genetic analysis of early flowering mutants in Arabidopsis defines a class of pleiotropic developmental regulator required for expression of the flowering-time switch flowering locus C.

The Arabidopsis flowering-repressor gene FLOWERING LOCUS C (FLC) is a developmental switch used to trigger floral induction after extended growth in the cold, a process termed vernalization. In vernalized plants, FLC becomes transcriptionally silenced through a process that involves an epigenetic mechanism. We identified recessive mutations designated vernalization independence (vip) that confe...

Vernalization - a cold-induced epigenetic switch.

Growth and development are modulated by environmental signals in many organisms. These signals are often perceived at one stage and 'remembered' until later in development. An increasingly well-understood example of this process in plants is provided by vernalization, which refers to the acquisition of the ability to flower after prolonged exposure to cold. In Arabidopsis thaliana, vernalizatio...

Nuclear organization changes and the epigenetic silencing of FLC during vernalization.

Changes in nuclear organization are considered an important complement to trans-acting factors, histone modifications and non-coding RNAs in robust and stable epigenetic silencing. However, how these multiple layers interconnect mechanistically to reinforce each other's activity is still unclear. A system providing long timescales facilitating analysis of these interconnections is vernalization...

Coordination of the vernalization response through a VIN3 and FLC gene family regulatory network in Arabidopsis.

Vernalization is an environmentally induced epigenetic switch in which winter cold triggers epigenetic silencing of floral repressors and thus provides competence to flower in spring. Vernalization triggers the recruitment of chromatin-modifying complexes to a clade of flowering repressors that are epigenetically silenced via chromatin modifications. In Arabidopsis thaliana, VERNALIZATION INSEN...
